6. Protect label identity through every batch change

Confirm which label belongs to each product and pack size, where the batch number and expiry information will be applied, and how their presence and legibility will be checked. Define line clearance, removal of previous-batch materials, label reconciliation where applicable, and documented release before restarting.

For US finished-drug manufacturing, 21 CFR 211.130 addresses packaging and labeling controls, including prevention of mix-ups and documented pre-use inspection. A bottle labeling machine applies labels; coding, vision inspection, serialization, aggregation and electronic records require their own specified hardware, software and verification scope.

7. Turn the URS into an acceptance plan

Write the User Requirements Specification (URS) before the configuration is frozen. Define what the equipment must do, how it will be cleaned and operated, and the evidence needed for acceptance. Assign responsibility for utilities, upstream and downstream interfaces, testing, deviations and document approval.

Factory Acceptance Testing (FAT) checks the agreed configuration at the supplier’s premises; Site Acceptance Testing (SAT) checks it after installation and integration. IQ documents installation against the approved specification, OQ checks operation across defined ranges, and PQ demonstrates performance under the intended production conditions. These activities support an agreed qualification and validation strategy; FAT alone does not validate the pharmaceutical manufacturing process. Start with the dosage form, formulation properties and package. Oral liquids require a suitable pump and nozzle; ointments need compatible tube filling and sealing; powders or granules in hard capsules need a capsule filler; finished tablets and capsules need a counter for bottle packing. Confirm suitability with samples and an agreed test plan.

Multiple formats can be included when their dimensions and operating requirements are assessed during design. Agree the change parts, guides, pucks, tube holders or capsule tooling needed for each format, along with setup and cleaning procedures. Compatibility should be checked for every proposed format.

Define the target volume, capsule fill weight or bottle count together with allowable deviations and the sampling method. Confirm the formulation or justified test medium and the intended operating speed. Published figures describe a particular model or condition; agreed line output must account for every integrated operation.

Review the complete contact path, residue risks, access for dismantling, tubing and seal replacement, and the intended cleaning agents. Establish procedures and records appropriate to the pharmaceutical process. CIP and SIP are configuration-dependent and must be specified; they are not standard capabilities of every machine.

Provide the dosage form and relevant formulation properties, packaging drawings and representative samples, fill volume or count, output target and available layout and utilities. Include cleaning, environmental, inspection and document requirements. Agree sample-handling and trial arrangements before sending product.

Equipment can support a defined pharmaceutical process, but GMP compliance depends on the installed system, quality procedures, trained personnel, maintenance and appropriate qualification and validation. Agree the supplied documentation and testing support with GDHP; the pharmaceutical manufacturer retains responsibility for quality approval and regulatory compliance.
